Robin van Persie and Nemanja Vidic won't be joining the team for tonight's match as they are both injured. The pair also missed United's 2-2 draw against Cardiff.
As well as those injuries, midfielder Marouane Fellaini is missing tonight's match due to suspension.
A win tonight is vital for both teams as it would ensure a place in the final 16 but a draw could suffice if Shakhtar Donetsk lose to Real Sociedad, another game on the group.,
The manager of Manchester United, David Moyes, said about Van Persie and Vidic: "They are both injured. Nemanja Vidic had concussion and Robin has a problem with his groin. Neither of them are long-term but they are not quite ready yet."
Other than United's last minute loss against Cardiff on Sunday the team has a smashing 10-game unbeaten run at the moment and are confident going into tonight's game.
For their part Bayer Leverkusen are also not starting with a full line up of their best players. Winger Sidney Sam, who injured his thigh in a game on Saturday, will not be playing tonight.
Sami Hyypia, the manager of Bayer Leverkusen noted that since his team lost to Manchester United 4-2 in the group's opening round they picked up seven points in their next three games.
"We have always played well at home," said Hyypia, adding: "It won't be easy for Manchester United and, of course, we really want to win."
